No, the well will stay primed, but what Blue is referring to is when it is freezing outside and you wake up and the well lost prime for whatever reason: loss of electicity or the lines froze up, and you have to go out there freezing your "****" off to prime it.
Wells never seem to lose prime on warm sunny days!
